On 5 Apr 1675, a year after the death of their father, siblings Alleyne Culpeper, Francis Culpepper and Margaret Culpeper Skeete were named in the will of their uncle, Reynold Alleyne. No other siblings were listed. On 19 Dec 1689, Alleyne witnessed the will of Thomas Rouse.
